Autonomous discovery, development and optimization of organocatalytic reactions through intelligent chemical robots
As a result of the recruitment process, the postdoctoral position has been offered to Dr Ahmad Kassir.
Chirality plays an important role in nature, as molecules of different chirality can exhibit vastly different properties. A large part of the field of chemical catalysis, and in particular of organocatalysis, is devoted to the selective synthesis of chiral molecules. However, despite the huge progress made in recent decades, we are still far from being able to efficiently design the synthesis of any chiral molecule. Usually, the development of new (organo)catalytic methods requires extensive and time-consuming optimization, which is usually performed manually. Therefore new enabling technologies are needed to speed up this process. Automation and robotic approaches have started to play an important role in the chemical sciences. The use of automated approaches can speed up the research process, make the experiments more repeatable and free researchers from repetitive tasks, so they can focus on the design of new transformations. In this project, we aim to investigate the field of organocatalysis with chemical automation and robotics that will ultimately be guided by artificial intelligence methods.
Description of tasks:
The successful Candidate will work on several goals:
– the design and construction of high-throughput automated robotic platform
– automated optimization of selected transformations
– autonomous searching for new transformations with robotic platform
– synthesis of starting materials, catalysts, and building blocks
– mechanistic investigation of selected transformations as well as probing their scope and limitations

Chiral hybrids of quinones and quinols with double anticancer and antimicrobial activity against drug-resistant pathogens as potential agents in hospitalization of cancer treatment.
Cancer is a crippling and challenging disease. Despite the progress in the development of cancer treatments, there is no suitable cure for most advanced cancers.
Cancer patients are also affected by multidrug resistant (MDR) infections, often associated with considerable morbidity and mortality. On the basis of the literature reports, naturally occurring quinones and quinols have been identified as groups of compounds with both of these activities.
Our preliminary results show that the decarboxylative coupling reaction is a convenient tool for the synthesis of variously substituted quinones, which can be further transformed to quinols. However, a stereoselective variant of this is available.
The reaction remains unknown. The main topic of the project is to develop a selective decarboxylative cross-coupling reaction using various aromatic and aliphatic chiral nonracemic acids. After optimisation, a compound library will be prepared and used for biological assays for antimicrobial and anticancer activity. The results obtained will be correlated with the dual inhibition properties towards enzymes recognised in cancer therapy, thioredoxin reductase 1 (TrxR1) and tyrosinase.
Anticancer cell assay and part of antimicrobial assays will be conducted in cooperation with our partners from the European Network Action COST CA21145 for the diagnosis and treatment of antibiotic-resistant bacterial infections (EURESTOP). The Molecular Knocking of selected compounds with enzymes will be carried out with our partner from the COST CA21162 action, Establishing a Pan-European Network on Computational Redesign of Enzymes (COZYME).
Description of tasks: Analysis of data collected in our team and analysis of literature data. Synthesis of chiral substrates. Performing catalytic decarboxylative coupling reactions. Preparation of reports and manuscripts.

Approaching 20% emission efficiency in the NIR-II region with radical chromophores
The overarching goal of this project is to uncover a strategy to create radicals which at the same time: (a) strongly emit light in the NIR-II region; (b) are stable under ambient conditions; (c) strongly absorb light; (d) display large circularly polarized luminescence. The primary objective of ARCHIMEDES is to deliver breakthrough organic materials possessing large fluorescence quantum yields and stable radical structures in the integrated fields of molecular design, chromophore synthesis and fluorescence imaging of living cells. The realization of ARCHIMEDES will be based on both expanding the chemical space of stable, emissive C-centered radicals and on heretofore nonexisting emissive nitroxide radicals. The synergistic effects of increased brightness of NIR-II dyes and the higher sensitivity and resolution offered by CPL fluorophores will provide quality fluorescence imaging on a previously unthinkable level.

Stereoselective dearomatization of nonactivated arenes via an „alkene walk” pathway: Rapid access to high-added value poly- and spirocyclic systems from readily available aromatic compounds.
The aim of this research proposal is to establish a new methodology for the stereoselective dearomatization of nonactivated arenes via the virtually unexplored retro-ene reaction of in situ generated benzylic diazenes (a so called “alkene walk” process), and subsequent transformation of the so formed active polyene species into chiral poly- and spirocyclic molecules. As a result, a new and versatile tool for the synthesis of high-value organic molecules from cheap and readily available benzylic alcohols, halides, aldehydes (in combination with organometallics) and monosilyl diazenes will be added to the organic chemist’s toolbox. Moreover, the possibility of synthesis of advanced polyaromatic systems via an additional re-aromatization process will be tested.
The candidate will be responsible for synthesizing of the starting materials and polyene intermediates, followed by an studies on their selective of transformations into variety of poly- and spirocyclic systems using selected literature protocols. The candidate will also be responsible for recording and interpreting spectral data, designing experiments, and preparing reports and materials for publications. Complementary theoretical studies on reaction mechanisms may be performed by the candidate or as part of a collaboration, depending on the scholar’s interests.

“Self- or a directing group-aided remote C(sp3)-H functionalization of difunctional compounds”
The announced research project, funded by Polish National Science Centre, is devoted to develop novel methods for self- and a directing group aided C(sp3)-H functionalization of simple bi- and polyfunctional organic molecules. Most of the efforts will be put on the development mild and versatile protocols under modern radical photoredox or electrochemical conditions. Previous experience with organic photochemistry and/or electrochemistry will be an additional asset.
The PhD candidate (PDC) will be responsible for synthesizing, depositing, and characterizing of the starting materials and intermediates, followed by an optimization of reaction conditions for model transformations of radical initiated remote C(sp3)-H functionalization of bi- and polyfunctional organic compounds. In extension, the PDC will investigate a scope of the developed protocols, including starting material variations, reaction condition variation (photoredox, electrochemical, transition-metal catalysed process). Key efforts will be put on an investigation of asymmetric mode of the investigated C-H functionalization along with application studies devoted to synthesis of the selected bioactive or medicinally important molecule by using investigated process. Complementary theoretical studies on reaction mechanisms may be performed by the PDS or as part of a collaboration, depending on the scholar’s interests. To a certain degree, the PDC will be given intellectual freedom in defining their research.
The PhD candidate will also participate in the Institute scientific life by attending to group and institute seminars and lectures. The PhD candidate will also attend to advanced course devoted to modern organic chemistry and spectroscopy.

“Chemoenzymatic cascades of new Cu and Pd reactions of significant application potential”
As a result of recruitment for the PhD student position, no candidates were selected
Currently, an increase in the incidence of cancer is observed, thus it is extremely important to eliminate or limit them. One of types of cancer is multidrug resistant (MDR) solid tumors.. The key challenge of this action is an identification of a new diagnostic/predictive biomarkers, producing new and safe compounds applicable to personalized treatments of MDR tumors. In order to achieve such goal, we need to design new synthetic protocols providing organic compounds of defined structure, free from any impurities. Therefore, during the realization of project we will study a new type of organic reactions catalyzed by copper salts, which provide molecules of high activity towards MDR. To achieve success, we need to know reaction scope and limitations, reaction mechanisms and optimal conditions for successful synthesis. Since the obtained products will be applied for biological and medical studies by COST specialists, they should be free from impurities including residual metal catalyst. Therefore, in the project special attention will be paid on reaction conditions and catalyst immobilization/inclusion to design experimental procedure providing metal free products.
Description of tasks: Analysis of data collected in our team and analysis of literature data. Synthesis of catalysts. Performing catalytic reactions with copper-based catalysts and discovery new catalytic reactions based on data analysis. Preparation of reports and manuscripts.

“Chasing lithium: direct lithium salt extraction and purification from mixtures using stimuli-responsive “smart” molecular extractants”
As a result of recruitment for the PhD student position, the following were employed: Jakub Narodowiec
Motivation: The rapid development of the electrotechnical industry has contributed to a dramatic growth in demand for lithium, which is used primarily in Li-ion batteries. This element is crucial for shifting our society from burning fossil fuels to clean energy generated from renewable sources. Considering the strategic interest in this most valuable resource of the 21st century, identifying and developing the practical and most sustainable lithium supply and recycling strategy is crucial for a successful transition to a low-carbon economy. Lithium is extracted primarily from minerals and brines and, to a lesser extent, from spent Li-ion batteries. In these resources, lithium typically co-occurs with various metal salts (e.g., sodium, potassium, magnesium, and calcium). The similarity between the associated salts (e.g., NaCl vs. LiCl) renders selective extraction of lithium a challenging and resource-consuming task. In addition, alternative potentially competing metal cations, such as magnesium and calcium, featuring higher specific charge densities often make selective lithium binding even more challenging. However, the current methods have significant limitations, such as tremendous energy requirements or the need to use large amounts of water and toxic reactants, resulting in a massive environmental impact. Thus, developing sustainable lithium extraction and purification technologies is one of the key research directions. Studies toward using synthetic molecular receptors containing cation and anion binding sites in their structure is an appealing solution to achieve direct lithium extraction (DLE) from the mixtures. However, the current molecular extractants have some limitations, such as inefficient synthesis, poor lithium selectivity, and slow kinetics of lithium salt release due to the high stability of the formed complex. In other words, lithium extraction using these systems can take from hours to days. Consequently, these systems are not suitable for practical use.
The aim of the research: In this project, we will evaluate the feasibility of employing photoswitchable receptors as direct extractants and transporters of lithium salts (Li-salts) of industrial importance, such as lithium chloride (LiCl). In principle, these “smart” systems exhibit capability for directly extracting Li-salts from the mixtures of various competing solid salts and their aqueous solutions to organic solvents, from which Li-salt can be recovered in pure form. The proposed azobenzene-based electroneutral extractants are equipped with lithium-cation-selective binding groups and anion binding groups selected for particular anions such as chloride.
We expect that the native trans-form of the extractant will demonstrate a weak affinity for Li-salts due to the spatial separation of cation and anion binding domains. In contrast, the light-generated metastable cis-form of the extractant will show superior binding, extraction, and transport of Li-salt due to the proximity and advantageous arrangement of ion-binding domains, shielding the guest from the environment. The efficient bidirectional trans/cis isomerization of the robust azobenzene linker allows control of the complex’s thermodynamic stability and the rate of catching and releasing Li-salt from the host binding cavity. All these features will enable efficient and energy-efficient management of the Li-salt binding and release process, allowing the development of the continuous extraction of Li-salt from the mixtures for the first time.
The project is divided into sections. Firstly, we will initiate the theoretical studies to predict the molecular properties of the proposed trans/cis-extractants and their putative complexes with various salts, such as chlorides of alkali metal cations. We aim to synthesize only the compounds predicted to display selectivity for lithium over competing cations. Then, for chosen systems, we will examine in detail their solution-binding properties and extraction efficiency with more time-consuming 1H/7Li NMR titration techniques and solid-liquid- (SLE) and liquid-liquid extraction (LLE) studies. The theoretical model will be tuned due the course of the project by comparison with the experimental data from the binding and extraction studies. Finally, selected hosts will be tested continuously as the first stimuli-responsive lithium salt extractants using a custom-made testing apparatus.

“Stereoselective dearomatization of nonactivated arenes via an „alkene walk” pathway: Rapid access to high-added value poly- and spirocyclic systems from readily available aromatic compounds”
As a result of recruitment for the PhD student position, the following were employed: Rajasekar Marimuthu
Stereoselective dearomatizations, especially in non-activated arenes, allowing conversion of readily available planar aromatic compounds into three-dimensionally complex molecules with precise control of reaction site, regio- and stereoselectivity has long been an exciting challenge in organic synthesis. Due to their high resonance stabilization energy, dearomatizations of nonactivated arenes usually require the use of stoichiometric amounts of expensive and toxic transition metal activators or elevated temperatures and harsh reaction conditions. This makes achieving high stereoselectivity in these processes very difficult. In our approach aromatic substrates are initially transformed into highly active polyenes which can then further derivatized using the broad spectrum of well-established stereoselective reactions of alkenes and dienes conducted under mild reaction conditions, resulting in highly stereoselective dearomatizations. Further advantages of the method include high availability of substrates (in the case of chiral derivatives in both enantiomeric forms in high optical purity) and high chemo- and stereodiversity of the obtained products.
The PhD candidate will also participate in the Institute scientific life by attending to group and institute seminars and lectures. The PhD candidate will also attend to advanced course devoted to modern organic chemistry and spectroscopy.

“Artificial intelligence-driven design and discovery of chiral phosphoric acid catalysed reactions”
W wyniku rekrutacji na stanowisko post-doc zatrudniono: dr Chandrakanta Parida
Assymetric synthesis is one of the most important branches of contemporary organic chemistry as synthesis of chiral compounds is of great interest in the fields of materials science, agrochemicals and pharmaceutical industry. However, the discovery of novel asymmetric transformations is usually time-consuming and relies in many cases on serendipity. The artificial methods promise to speed up the navigation of chemical space, but little is known how to design new transformations from first principles. Deep learning was successfully applied to many fields achieving super-human performance, yet its full potential in chemistry is to be achieved. The current project investigates the application of machine learning methods and the successful candidate will be involved in discovery and optimization of new catalytic reactions. The work will include organic synthesis of starting materials and reagents, as well as substrate scope investigation, products structure determination, formulation of research hypotheses, analysis and interpretation of obtained results.

“Chemoenzymatic cascades of new Cu and Pd reactions of significant application potential”
Currently, an increase in the incidence of cancer is observed, thus it is extremely important to eliminate or limit them. One of types of cancer is multidrug resistant (MDR) solid tumors. This is the subject of COST action CA17104.
Performing catalytic reactions with
copper-based catalysts and discovery new catalytic reactions based on data analysis. Preparation of reports and manuscripts.

“Autonomous discovery, development and optimization of organocatalytic reactions through intelligent chemical robots”

Performing catalytic reactions with
copper-based catalysts and discovery new catalytic reactions based on data analysis. Preparation of reports and manuscripts.

“Self- or a directing group-aided remote C(sp3)-H functionalization of difunctional compounds”
The announced research project, funded by Polish National Science Center, is devoted to develop novel methods for self- and a directing group aided C(sp3)-H functionalization of simple bi- and polyfunctional organic molecules. Most of the efforts will be put on the development mild and versatile protocols under modern radical photoredox or electrochemical conditions. Previous experience with organic photochemistry and/or electrochemistry will be an additional asset.
The PhD candidate (PDC) will be responsible for synthesizing, depositing, and characterizing of the starting materials and intermediates, followed by an optimization of reaction conditions for model transformations of radical initiated remote C(sp3)-H functionalization of bi- and polyfuntional organic compounds. In extension, the PDC will investigate a scope of the developed protocols, including starting material variations, reaction condition variation (photoredox, electrochemical, transition-metal catalyzed process).

“CO2CHEM – redox-neutral photocatalytic c-h carboxylation of hydrocarbons with co2“
The general goal of this proposal is to develop the synthetic and mechanistic basis for the photocatalytic, redox-neutral C-H carboxylation of saturated hydrocarbons with CO2. To address this challenge, we propose new synthetic strategies based on photocatalytically generated alkyl radicals and earth-abundant metal complexes to provide C-H carboxylation in an overall redox-neutral process.
The project will be realized in strong collaboration with prof. B König from the Faculty of Chemistry and Pharmacy, University of Regensburg, Germany.
In particular, the main tasks will involve:
– design and synthesis of cobalt-complexes and their evaluation in photo-organo-metal catalysed C-H carbonylation with CO generated in situ;
– investigations on the photo-organo-metal catalysed C-H carbonylation with CO generated in situ;
– mechanistic investigation on the most interesting C-H carboxylations;
– scope and limitation studies;
– preparation of manuscripts.

“Sustainable, photochemical transformations with diazo reagents as a source of reactive intermediates”
Applications are invited for a post-doc position in a research grant “Sustainable, photochemical transformations with diazo reagents as a source of reactive intermediates” funded by the National Science Centre, Poland. Successful candidates will work in the Sustainable Chemistry Group under the supervision of Prof. Dorota Gryko.
Drawing inspiration from photosynthesis, we challenged the use of visible light for the activation of organic molecules and the discovery unknown solar-driven transformations. Plethora of organic compounds have been investigated in terms of their photochemical activity. The fact that diazo compounds possess their intrinsic chromophore (-C=N+=N–) puts them in a unique position among many reagents that can absorb visible light and hence are suitable for solar-driven transformations.
The main objective of this proposal is to unveil general reactivity modes of diazo compounds under visible light-irradiation. We will develop their solar-driven reactions and determine reactive intermediates (singlet or triplet carbenes, radicals, others) generated during these processes.

“Synthesis of novel class of organic emitters for TADF OLED materials embracing curved nanographene fragments”
Lider XI project
1 (12 months)
R2
In this project, we focus on the rational design and synthesis of innovative organic architectures – nanographenes, (consisting of D-π-A structure) with positive or negative Gaussian curvature. Furthermore, the goal of the project is to develop an efficient and inexpensive methodology for nanographenes synthesis. In addition, great emphasis will be put on studying the correlation between electronic structure of nanographenes and their photophysical properties, using theoretical (DFT calculations) as well as experimental (NMR, UV-vis-NIR, emission, CV) approaches. Advanced low-temperature photophysical measurements will be then performed to identify compounds with the highest photoactivity in both, solution and solid state. Nanographenes will be employed in ultimate task to fabricate TADF OLED devices and identify their emissive throughput and stability.
Key responsibilities include:

In recent years a novel circuit element called memristor is introduced whose conductivity in present depends on the history of electronic current passed through it in the past. A memristor is a device which can be used for in-memory processing, therefore, highly valuable for reducing the processing energy and elevating the processing speed by bypassing von Neumann bottleneck, i.e. CPU-memory data transfer. Currently, most of the known memristors are based on balk materials, often oxides of metals. A few molecular memristors are known which are either slow to respond, built on the basis of precious metals, or too unlikely to be fabricated due to complicated preparation processes. We aim to design, synthesize, and perform measurements on all-organic molecular memristors. A notable part of the design is already taken care of. However, still a lot is remained to be explored. In particular, we need to perform non-equilibrium Green’s function computations to compare transmission properties of a huge number of molecules in detail. The successful candidate will work in close collaboration with a dynamic team of organic chemists, theoretical chemists, and solid-state physicists. In our group you witness how the molecules you make, can make a difference in human life.
